Description
The preparative small-scale production of non-protein amino acids was created in 2007 with the financial support of the International Science and Technology Center (ISTC). As a result of scientific research, a universal technology has been developed that allows obtaining important and expensive enantiomerically pure non-protein α-amino acids (market price of 1 g is $ 150-400) of various structures from inexpensive natural glycine amino acids. and D, L-alanine (the price of 1 kg is about $ 10). Using the developed technology, more than 200 new amino acids not described in the literature were synthesized that contain aliphatic, aromatic and heterocyclic substituents in the side chain.
As irreversible enzyme inhibitors, non-protein amino acids are of great interest in pharmacy, medicine, science and technology. To date, there are numerous, well-known anti-tumors, antiviral, antihypertensives and other drugs whose pharmacologically active aglycon is a non-protein amino acid or its derivative.
As chiral compounds, non-protein amino acids are suitable only for use as optically pure stereoisomers. There are many cases where the optical antipode of a pharmacologically active drug has a negative pharmacological effect or, at best, remains neutral. Therefore, to obtain such compounds, it is necessary to develop methods that provide optical stereoselective high enantiomeric purity. The technology of asymmetric synthesis, developed at the Scientific and Production Center “Armbiotechnology” of NAS RA, allows one to obtain amino acids with an enantiomeric excess (of it) of more than 98% regardless of the structure and degree of chirality. Modern physicochemical methods are used to determine the structure and absolute configuration of the synthesized new amino acids, such as 1 H and 13 C. NMR, chiral HPLC and GLC, LC-MS, elemental analysis, polarimetric measurements, etc. are used.
A small production line of non-protein amino acids has already been introduced at the ME C level and about 13 enantiomerically pure non-protein amino acids are produced by D and sold on a contractual basis. The main customers are Belgian ACROSS Organics and German IRIS Biotech GmbH.
